Matthews left the team's practice on Tuesday with an upper-body issue and Jarnkrok picked up an upper-body ailment in Ottawa.
The Toronto Maple Leafs will dress a lineup that should closely resemble their opening night lineup.
Toronto plays their third of six pre-season games on Thursday night against the Montreal Canadiens inside Scotiabank Arena. The lineup will feature mostly NHL players, including Easton Cowan, pushing to make the squad out of training camp.
Other notable players pushing for a spot in Toronto's lineup are Max Pacioretty and Steven Lorentz, both on professional tryouts with the club. Nick Robertson and Pontus Holmberg are also looking to make the team and will appear in their second straight pre-season game.
Toronto's lines will likely resemble how they were at practice on Wednesday afternoon.
Auston Matthews, who had a maintenance day on Wednesday after dealing with an upper-body injury a day earlier, won't play in Toronto on Thursday. Calle Jarnkrok isn't in the lineup either. He suffered an lower-body injury in Tuesday's game against the Ottawa Senators.
Both players were on the ice ahead of the Maple Leafs' optional morning skate on Thursday morning.
Maple Leafs head coach Craig Berube hinted that the lines ahead of this season will soon take shape.
"It's about time we get our team together and do things together," he said on Thursday afternoon. "Chemistry is important. Combinations are important."
Matthew Knies is one notable player absent from Thursday's roster. He could likely be out of the lineup due to the potential competition within the bottom six. He'll skate in the second group.
Matt Murray and Anthony Stolarz are the two netminders ahead of their third pre-season game.
Catch the game nationally on Sportsnet. Puck drop is scheduled for shortly after 7:00 p.m. ET.
